Velocity Sedimentation Analysis of Drug-Induced Oligomers of Tubulin

Each curve for a sample is an A280 scan (5 minute intervals) from the tops to bottoms of tubes after reaching 35,000 rpm (sample height at t0 ~1 cm).

Conclusions:
1) These drugs all induce tubulin oligomers that sediment more rapidly than tubulin alone.
2) The effects of the various drugs are distinct.
3) Mathematical analyses indicate the presence of two predominant, oligomeric species in the presence of hemiasterlin* and vinblastine.

* [Hemiasterlin] is substoichiometric.
After ~12 hours, only the 19.0 s species is detected (data not shown).
